Multidrug resistance in bacteria has become a threat to public health care over the world. One of the bacteria with a high rate of resistance in antibiotics is Escherichia coli. The most important resistance mechanism to antibiotics in E. coli is through a tripartite AcrAB-TolC pump system that belongs to the RND superfamily. AcrB plays a major role in this system. There are two binding pockets on the AcrB pump (proximal binding pocket and distal binding pocket) where subtracts bind and effect to AcrB. Therefore finding these pump inhibitors which can interfere with two binding pockets is imperative. Most EPIs cannot be used for a clinical trial because of high toxicity. Therefore docking is used as an initial screening method to find potential EPIs which low toxicity and can use for further experiments such as in vitro, in vivo, even if clinical trial.
